目的 探讨骨髓间充质干细胞源性微泡(BMSC-MV)修复大鼠早发性卵巢功能不全的自噬机制。方法 大鼠骨髓分离培养骨髓间充质干细胞;超速离心法从骨髓间充质干细胞培养液中分离微泡;腹腔注射顺铂溶液制备早发性卵巢功能不全(POI)模型,制备后3 d尾静脉取血ELISA检测血清雌二醇(E2)及卵泡刺激素(FSH);尾静脉注射BMSC-MV移植治疗POI大鼠模型,移植后28 d尾静脉取血ELISA检测E2、FSH及抗苗勒管激素(AMH),同时取卵巢组织检测自噬相关蛋白LC3及P62。结果 模型对照组及微泡移植组在模型制备后3 d的E2 含量低于正常对照组,FSH 含量高于正常对照组(P<0.001);微泡移植组在移植后28 d的E2、AMH含量高于模型对照组(P<0.001),FSH含量低于模型对照组(P<0.001);微泡移植组的LC3较模型对照组表达升高,而P62表达降低(P<0.001)。结论 BMSC-MV介导自噬修复大鼠早发性卵巢功能不全。
Objective To investigate the autophagy mechanism of bone marrow mesenchymal stem cell-derived microvesicle (BMSC-MV) in repairing premature ovarian dysfunction in rats. Methods The whole bone marrow adherence method was used to isolate,culture and identify BMSCs of SD rats. Microvesicles were isolated from bone marrow mesenchymal stem cell by ultracentrifugation. Premature ovarian insufficiency (POI) model was prepared by intraperitoneal injection of cisplatin solution,and serum estradiol (E2) and follicular stimulating hormone (FSH) were detected by ELISA from tail vein 3 days after preparation. Rat model of POI was treated with BMSC-MV transplantation by tail vein. Blood from tail vein was collected 28 days after transplantation to detect E2,FSH and AMH by ELISA. Meanwhile,ovarian tissues were collected to detect autophagy-related proteins LC3 and P62. Results The E2 content of the model control group and the microvesicle transplantation group was lower than that of the normal control group,and the FSH content was higher than that of the normal control group (P<0.001). The content of E2 and AMH in the microvesicle transplantation group at 28 days after transplantation was higher than that in the model control group (P<0.001),and the content of FSH was lower than that in the model control group (P<0.001). Compared with the model control group,LC3 expression in the microvesicle transplantation group was increased,while P62 expression was decreased (P<0.001). Conclusion BMSC -MV mediate autophagy to repair premature ovarian insufficiency in rats.
目的 探讨臭氧水在外科手消毒中的消毒效果和医护人员的舒适度。方法 将参加手术的医务人员80例按时间段分为对照组、低浓度组、高浓度组,于消毒后分别进行手采样培养,并进行分析。结果 消毒后三组在菌落数比较差异有统计学意义(P<0.01);消毒后三组皮肤在发痒、脱屑、干燥评分比较差异有统计学意义(P<0.05);在气味、粘腻感、干燥速度比较差异有统计学意义(P<0.05)。结论 臭氧水用于外科手消毒效果可靠,无刺激气味和粘腻感,医护人员感觉更舒适。
Objective The aim of this study was to explore the disinfection effect of ozone water in surgical hand disinfection and the comfort of medical staffs. Methods 80 medical staffs who participated in surgeries were divided into control group, low-concentration group and high-concentration group according to the time period. After disinfection, hand samples were cultured and analyzed. Results After disinfection, there were statistically significance in colony number (P<0.01), itching, scaling and drying scores (P<0.05), odor, stickiness and drying speed (P<0.05). Conclusion Ozone water for surgical hand disinfection is reliable, no irritating odor and stickiness, and makes medical staffs feel more comfortable.
目的 运用网络药理学方法预测生白术活性成分、作用靶点及生物学意义,探讨其防治便秘的作用机制,并结合导师临床应用取得的疗效进行进一步的验证。方法 借助TCMSP在线数据库查找白术的药效成份并选择其生物利用度(OB)>30%且类药性(DL)>0.18的化合物,并查询每种成分所对应的靶标。通过Gene Cards、OMIM共2个疾病相关靶点的数据库检索便秘相关靶点信息。将二者靶基因相映射获得交集靶点。借助 cytoscape 3.7.1 软件对查询结果进行可视化。所得到的基因通过相互作用数据库(STRING)进行相互作用蛋白查询并构建蛋白质相互作用(PPI)网络。使用R语言对关键靶点行GO和KEGG富集分析,以构建“成分-靶点-信号通路”的网络。结果 共得到白术人源靶蛋白7个,便秘相关的人源基因2 859个。发现其主要通过干预PGR、CHRM3、CHRM1、ACHE、CHRM2五个基因并参与胆碱能突触、钙信号通路、肌动蛋白细胞骨架的调控、神经活性配体-受体相互作用、cAMP信号通路、PI3K-AKT信号通路共6条信号通路以达到防治便秘的效果。结论 应用网络药理学方法分析预测得到重用生白术防治便秘的潜在药效成分、作用靶点及其信号通路,为临床应用提供了理论依据。
Objective To predict the active ingredients, targets and biological significance of Atractylodes macrocephala by network pharmacology, to explore the mechanism of its prevention and treatment of constipation, and to further verify its efficacy in combination with the clinical application of tutors. Methods The constituents of Atractylodes macrocephala were searched by TCMSP database and the compounds with bioavailability (OB) > 30% and drug-like property (DL) > 0.18 were screened, and the corresponding targets of each constituent were queried. Constipation-related target information was retrieved from two disease-related target databases of GeneCards and OMIM, mapping the two target genes to obtain intersecting targets, by visualization of query results with cytoscape 3.7.1. The resulting genes were queried by the interaction database (STRING) and the protein interaction (PPI) network was constructed. GO and KEGG enrichment analysis of key targets was carried out by R language in order to construct the network of “component-target-signal pathway”. Results Seven human target proteins and 2 859 constipation related human genes were obtained from Atractylodes macrocephala. It was found that the effect of prevention and treatment of constipation was mainly achieved by interfering with five genes of PGR, CHRM3, CHRM1, ACHE and CHRM2 and participating in six signaling pathways: cholinergic synapse, calcium signaling pathway, regulation of actin cytoskeleton, neuroactive ligand-receptor interaction, cAMP signaling pathway and PI3K-AKT signaling pathway. Conclusion The potential pharmacodynamic components, targets and signaling pathways of reuse Rhizoma atractylodis macrocephalae in the prevention and treatment of constipation can be predicted by network pharmacological method, which provides a theoretical basis for clinical application.
目的 对下颌角肥大钻凿法截骨整形手术进行生物力学研究,并探索该术式的优化改进。方法 采用有限元方法,建立下颌角钻凿法截骨整形手术及其改良术式的三维有限元模型并进行应力分析、比较。结果 模型-1(钻凿法下颌角截骨整形手术模型)下颌角断裂失效载荷在所有术式中最高,为2426N。其余的改良术式模型(模型-2~7)失效载荷均低于模型-1,分别为2018N、1854N、1447N、1183N、1028N、876N。同时,模型-7与其他模型对比,其应力分布范围最小,关节盘von Mises应力手术侧大于非手术侧,大部分区域为0.52~3.25 MPa之间,最大不超过9.86 MPa。结论 在非均匀钻孔加去下颌角颊侧皮质骨的同时施加最大咬合力和下颌支后缘约束的手术方法最优。
Objective To study the biomechanics of osteotomy surgery for mandibular angle hypertrophy with drilling method and to explore the optimization and improvement of this operation. Methods Using the finite element method, the three-dimensional finite element models of osteotomy surgery for mandibular angle hypertrophy with drilling method as well as its modified operations are established,meanwhile, the stress analysis and comparison are performed. Results Model-1 (mandibular angle osteotomy surgery model by drilling method) had the highest fracture failure load among all surgical methods, which was 2426N. The failure loads of the other modified surgical models (model-2 ~7) were all lower than that of model-1, which were 2018N, 1854N, 1447N, 1183N, 1028N and 876N, respectively. Meanwhile, compared with other models, the stress distribution region of model-7 was the smallest, with the von Mises stress in operating side of the articular disc was greater than that of the non-operating side, and most of the area was between 0.52~3.25MPa and the maximum of 9.86MPa. Conclusion The non-uniform drilling and removal of the buccal cortical bone of the mandibular angle are combined with the maximum occlusal force and the constraint of the posterior margin of the mandibular ramus is the best operation method.
目的 探讨尿清舒颗粒治疗泌尿系统感染伴有尿路结石的疗效。方法 选取我院2015年1月—2017年1月收治的80例泌尿系统感染伴有尿路结石患者纳入研究对象,以随机数表法分为观察组与对照组,各40例。对照组静脉滴注亚胺培南-西司他丁进行治疗,观察组在对照组基础上加服尿清舒颗粒。对比两组结石排出疗效及治疗前后尿白细胞镜检、中段尿液的普通细菌和真菌培养数量,尿频、尿急、尿痛、血尿、排尿不尽、腰酸乏力评分各项指标。结果 观察组结石排出疗效优于对照组差异有统计学意义(P<0.05);观察组治疗后尿白细胞镜检与治疗前及对照组治疗后对比下降、两组治疗后中段尿液的普通细菌和真菌培养数量与治疗前对比降低,差异有统计学意义(P<0.05);观察组治疗后尿频、尿急、尿痛、血尿、排尿不尽、腰酸乏力评分与治疗前及对照组治疗后对比降低,差异有统计学意义(P<0.05)。结论 尿清舒颗粒治疗泌尿系统感染伴有尿路结石可有效发挥抗菌作用,排出结石,缓解患者临床症状,且安全性较高,具有较高的临床应用价值。
目的 探究血清外泌体对小鼠烫伤伤口愈合的促进作用及机制。方法 选取我院60只NIH小鼠作为研究对象,对其血清中的外泌体进行提取和分离,建立起皮肤烫伤模型。采用血清外泌体进行治疗,评估治疗的效果,观察皮肤组织的病理变化,检测小鼠成纤维细胞增殖和表皮HaCat细胞迁移使用血清外泌体受到的影响。结果 空白对照组和血清外泌体小鼠烫伤创口愈合时间比较,差异有统计学意义(P<0.05);血清外泌体作用24 h后,不同剂量OD值差异有统计学意义(P<0.05);10 μL Exo组和PBS组内部不同时间点OD值差异均有统计学意义(P<0.05);外泌体组和PBS组24 h的HaCat细胞迁移率相比较,差异有统计学意义(P<0.05)。结论 血清外泌体可以促进小鼠烫伤伤口的愈合,对成纤维细胞的增殖和表皮HaCat细胞的迁移作用也比较显著,可能成为临床治疗烫伤的新手段。
目的 探讨利用微信群保留机采血小板无偿献血者的应用效果。方法 从本血站穿越安全输血标准化管理系统中调取2015年1月1日—2018年12月31日捐献机采血小板无偿献血者信息,根据初次捐献机采血小板后是否自愿加入机采微信群的机采血小板无偿献血者分为实验组和对照组,统计分析各组机采血小板无偿献血者重复献血 2~3 次和≥4 次以上机采血小板无偿献血者人数及平均献血次数。结果 2015—2018年间机采血小板采集量、献血人数及平均献血次数逐年上升;重复献血 2~3 次和≥4 次以上人数,实验组均高于对照组,机采血小板平均献血次数实验组均高于对照组,其中2016年和2018年实验组机采血小板平均献血次数高于对照组(P<0.05)。结论 通过机采微信群预约与管理机采血小板无偿献血者可提高机采血小板招募的工作效率,有效扩大和稳定机采血小板无偿献血者固定队伍。
目的 探讨血必净注射液对SAP大鼠TLR4信号通路介导肠黏膜屏障功能障碍的影响。方法 24只SD大鼠随机分成空白组(n=8)、对照组(n=8)和治疗组(n=8)。对照组和治疗组用4.5%牛磺胆酸钠溶液胆胰管逆行注射制备SAP模型,空白组采用等量生理盐水逆行注射。治疗组在造模3 h后经鼠尾静脉注射血必净注射液(3 mL/kg)。三组大鼠造模后观察24 h,然后处死取胰腺和小肠组织送病理检查,采用荧光RT-PCR技术检测TLR4和NF-κB表达水平,采用ELSIA法检测血清TNF-α、IL-6、淀粉酶(AMS)及二胺氧化酶(DAO)水平,比较三组大鼠各项指标。结果 对照组和治疗组小肠组织TLR4和NF-κB表达以及血清TNF-α、IL-6、AMS及DAO水平均高于空白组(P>0.05),治疗组小肠组织TLR4和NF-κB表达以及血清TNF-α、IL-6、AMS及DAO水平低于对照组(P<0.05)。结论 血必净注射液通过干预SAP大鼠TLR4信号通路,降低小肠组织TLR4和NF-κB的表达,减轻小肠组织的炎症反应,对肠黏膜屏障具有一定的保护作用。
Objective To investigate the effect on intestinal mucosal barrier dysfunction (IBF) of Xuebijing injection mediated by Toll-like receptor 4 (TLR4) signal pathway in rats of severe acute pancreatitis (SAP). Methods 24 Sprague Dawley (SD) rats were randomly divided into Sham group (n=8), control group (n=8) and treatment group (n=8). The SAP model was established by retrograde injection of 4.5% sodium taurocholate into the biliopancreatic duct in control group and treatment group, while control group was injected with the same amount of saline. In treatment group, Xuebijing injection (3 mL/kg) was injected via tail vein after 3h of modeling. All rats were monitored and sacrificed after 24 hours of modeling. Samples of pancreas and intestine were collected for pathologic determination. A fluorescent RT-PCR was used to determine the expression of TLR4 and NF-κB of small intestine. The serum levels of TNF-α, IL-6, amylase (AMS) and diamine oxidase (DAO) were measured by using ELISA. All parameters of three groups were compared. Results The expression of TLR4 and NF-κB of small intestine in control group and treatment group were higher than it in control group (P<0.05), as well as the serum levels of TNF-α, IL-6, AMS and DAO (P<0.05). The expression of TLR4 and NF-κB of small intestine in treatment group were lower than it in control group (P<0.05), as well as the serum levels of TNF-α, IL-6, AMS and DAO (P<0.05). Conclusion Xuebijing injection may not only reduce the expression of TLR4 and NF-κB of small intestine, but also alleviate the inflammation reaction of small intestine by interfering with TLR4 signal pathway, which may have a protective effect on intestinal mucosal barrier in SAP rats.
瘦素(leptin)是由控制各种生理过程的脂肪组织合成和分泌的一类激素,通过作用于靶细胞膜上的瘦素受体并经信号传导在各器官和系统中发挥一系列生物学效应。肾脏是高血压常见的靶器官之一。相关研究表明,瘦素在高血压肾损害中发挥作用,其机制可能与氧化应激及其炎症反应有关。本文以瘦素对高血压肾损害及其相关机制作一综述,并探讨瘦素对高血压肾损害发病机制研究进展。
Leptin which is a kind of synthesis and secretion of hormone that participates in various physiological processes is the role of the leptin receptor on the target cell membrane and the signal transduction through a series of biological effects in different organs and systems. Kidney is one of the common target organs of hypertension, and related research shows that leptin plays a role in hypertensive kidney damage, whose mechanism may be related to oxidative stress and its inflammatory reaction. The paper reviewed leptin on renal damage in hypertension and its related mechanisms, to explore the leptin on renal pathogenesis of hypertension research progress.
目的 了解长沙市雨花区环卫工人的健康状况,为有针对性地制定健康教育措施提供依据。方法 为环卫工人进行体格检查,对其体检资料进行统计分析。结果 1 672名受检环卫工人中仅有57人所检项目完全正常,异常检出率达96.60%,其中男性职工的总体检出率要高于女性职工(P<0.05);男性职工慢性咽炎、高血压、血脂异常、高尿酸血症、肾囊肿的检出率较女性职工高(P<0.05);而女性职工肾结石、甲状腺结节、痔疮的检出率要高于男性职工(P<0.05)。体检结果尚具有一定的年龄段分布规律,膝关节退变、超重和肥胖、慢性咽炎、脂肪肝、高尿酸血症、甲状腺结节等检出率差异有统计学意义(P<0.05);其中膝关节退变、高血压、前列腺增生等检出率随着年龄的增长而增加。结论 环卫工人的卫生保健意识薄弱,进一步促进改善他们的工作条件,并对其进行健康教育非常必要。
Objective To understand the health status of sanitation workers in Yuhua District of Changsha city, and to provide evidence for the establishment of health education measures. Methods The sanitation workers were examined by medical examination and their physical examination data were statistically analyzed. Results In the 1 672 subjects of sanitation workers there were only 57 people seized items completely nor -mal. Abnormal rate was 96.60%. The total examination rate of male workers was higher than that of female workers (P<0.05); morbidity of male workers in chronic pharyngitis, hypertension, dyslipidemia, hyperuricemia and renal cysts were higher than female workers (P<0.05); morbidity of the female workers in kidney stones, thyroid nodules, hemorrhoids, was higher than that of the male workers (P<0.05). The examination results had age distribution. There was statistically significant in knee joint degeneration, overweight and obesity, chronic pharyngitis, hyperuricemia, fatty liver, thyroid nodule (P<0.05); the knee joint degeneration, hyperten -sion, prostatic hyperplasia detection rate increased with age. Conclusion The sanitation workers' awareness of health care is weak. It is necessary to further improve their working conditions and to carry out health education for them.
